When a user fills out one of my forms, after hitting Submit they receive the Thank You message, and I have also set it up where they receive a reply email with a copy of their submission. Is there a way to set it up that after I review their form and complete their request I can click a button in the original emailed request I received and it will send an autoreply stating "Your request has been completed" or something along those lines?

You can set Reply to Email field inside of your email Notification to person's email who fills out your form. It is attached to your email field from your form.
You can then reply to your users and let them know that their request has been completed.
Or if you will edit their submission you can add one additional Autoreponder email to your form with your custom message "Your request has been completed" and you can use conditional logic to sent that email when you approve request made by your user.
For example you can add one additional filed to your form (checkbox field with option approved) and hide that filed so, it is not visible to your users. You will see hidden field when you edit submission.
Then you can set condition to send Approved Autoresponder email to your users if approved option is checked.
