I am trying to get my form to work so that when I receive a an inquiry via JotForm, all I have to do is click Reply and the Sender's Email autofills. How do I do this? Right now, it defaults to firstname.lastname@example.org.

All you need to do is add an email field to your form. This is to be used to collect the email address of your clients.
Then in your email settings, select your notification. And click on the recipients tab. You should see the name of your email field there.
So when a client submits your form you get sent a notification with the reply address of what the client entered in the email field.
