I wonder whether it could be possible to prompt users to authenticate (using username:password combinations from a listfile) before filling out a form to submit data?

That's possible with us but you'll have to define the username and password using our Conditional Logic Tool: Conditional Logic on Forms.
1. You can try single form: http://www.jotformpro.com/form/32967583823972 (Enter: Name and Codes). To show the submit button if the combination is correct.
2. You can try multi page form: http://www.jotformpro.com/form/32967955240968 (Enter: Name and Codes). To skip to certain start page if the combination is correct. To skip to the custom error page if the combination is incorrect.
You can add a twist to disable submitting the same combination twice by following our guide for: How to Set Form Limits Based on a Unique Question.
You can also use Submissions Verifier: http://apps.jotform.com/app/submission_verifier. Not that close to your preferences but would authenticate user's email validity.
