I am looking to make a form for people to signup to volunteer. Once we have a position filled, we don't need it anymore. How can I make it so each option can only be selected once?

Yes it is possible to achieve your requirement.
You may like to take a look at out "Gift Registry" widget which should help you. This will ensure that once a position is selected, it will not be allowed to be selected again by other users.
Another option is to make your question a "Unique Question" but it will not work in your case as the check box question cannot be set as unique question.
