My company will be sponsoring a contest, whereas, the public will have to vote for the winner. We would like for only 1 vote per voter. Can we have the poll set up to where the voter will have to sign up prior to voting? Also, we are lookong at 100 contestants.

You could have a registration form which redirects to your 'Poll' form when it gets submitted.

Another feature that you could use is to limit one form submittal per IP address, by using 'Strict Check' in the 'unique submission' options:
Unfortunately, at this time, we dont have the ability to limit a form to specfic number of submissions in total, but I know this is something the developers are working on. It is, however, hard to say exactly when we will see this feature will be enabled.
You could disable a form manually after 100 submissions, and have some text on the poll or registration form that says only the first 100 entries will be counted.
