Is there a way to sell an item in 3 different colors, $25/item or $100/5 items. Regardless of color combos. So,
2 blue + 3 red = $100
3 blue + 3 red = $125
3 blue + 2 red + 5 purple = $200
3 blue + 3 red + 6 purple = $250

You can do that with the product options, first, set up the product and then add the options, while adding the color as an option you should enable the "create sub-products" feature, here is also a guide that will help you doing so: https://www.jotform.com/help/264-Create-Sub-Products-Based-on-a-Product-Option

This allows you to do that for all one color. But if you choose multiple colors the price is not adjusted for the total # ordered.

I want to sell t-shirts, mix and match colors, for $25 each or $100 for a pack of 5. The pack of 5 can be different colors. So if they order 15 t-shirts (any color combo), the price will be $300. If they buy 16 t-shirts it'll be $325.

I'm afraid you cannot track how many items are selected and then change the price based on how many items are selected.
But, you could create different groups of products, like this:
I created two products:
-One that is $25 per product and you can select up to 4
-The second one that is $20 per product and you can select 5 only

If I have the total # of t-shirts ordered, is it possible to then a customer choose from 3 color options and force the total of the color options to equal the total number of shirts ordered?

I'm not quite sure if I understood your question if in case you want to create a custom pricing for each color so it adds in the total when you select it, you can do that by enabling Special Pricing in the color option. Is this what you're trying to achieve? The user will have to manually select the color and quantity of their choice.
